ES-ECs aretestedforexpressionofMarkersusingantibody,VE-cadherin(CD144,VE-cadherinAntibody,C-19,sc6458,SantaCruz);AF1002(R&DSystem)orCD31/PECAM-1(PurifiedRatAnti-MouseCD31,CatalogNo.553370,BD)byimmunofluorescencestainingorFACS.C57BL/6MousePrimaryAorticEndothelialCellsarenegativeforbacteria,yeast,fungiandmycoplasma.Cellscanbeexpandedfor3-6passagesatasplitratioof1:2underthecellcultureconditionsspecifiedby CellBiologics. Repeatedfreezingandthawingofcellsisnotrecommended.

LaboratoryApplications

ES-ECs canbeusedinassaysofcell-celladhesion,migration,vasculartubeformation,ortransendothelialresistance(TER).StandardbiochemicalproceduresperformedwithendothelialcellculturesincludeRT-PCR,Westernblotting,immunoprecipitation,immunofluorescentstaining,immunofluorescentflowcytometry,orgeneratingcellderivativesfordesiredresearchapplications.
